Home/ Applications / Gentle Mouse B Cell Enrichment to Maintain Cell He...
Are you struggling to isolate B cells in their native state? Are common processing methods damaging target cells or disturbing their physiology?
Akadeum’s revolutionary microbubble approach to B cell isolation delivers enriched B cells while maintaining cellular health and physiology. The isolated B cells retain their activation and differentiation potential throughout the enrichment process along with other defining characteristics such as their B cell receptor (BCR) isotype distribution, clearly indicating that the isolated populations are not being skewed or altered during the separation. The microbubble workflow is fast and easy to perform (simply mix to bind, spin to separate, and aspirate to discard) while being exceptionally gentle on delicate immune cells.
Analysis of B cell function and regulation necessitates a highly enriched population of unchanged B cells, but traditional separation techniques like magnetic bead-based sorting can be harsh on delicate immune cells and ultimately produce subpar results. Akadeum’s Buoyancy Activated Cell Sorting (BACS™) microbubble technology represents a breakthrough in next-generation cell isolation that allows for novel capture of unaltered cells in their natural physiological state.

Akadeum’s Mouse B Cell Isolation Kit delivers a highly enriched population of healthy, viable B cells using a protocol that is fast, easy, and exceptionally gentle on delicate immune cells.
Using Akadeum’s exceptionally gentle, microbubble-based approach to separation allows for the enriched B cells to retain their activation and differentiation potential. This novel, non-invasive cell isolation method does not disturb BCRs or skew the isolated populations.
BCR Isotype Distribution Remains Unchanged Following Microbubble Isolation
A research group at the University of California San Francisco recently used the Mouse B Cell Isolation Kit from Akadeum Life Sciences. Using the Akadeum Mouse B Cell Isolation Kit delivered an enriched population of B cells. Critically, the BCR percentages were similar in the enriched sample to the starting material, confirming that the isolation protocol didn’t disturb these receptors or skew the isolated population.
Antigen Presenting Capacity Retained After B Cell Enrichment with Microbubbles
A researcher from the Doug McNeel Lab at the University of Wisconsin recently used the Mouse B Cell Isolation Kit from Akadeum Life Sciences. The team was looking to obtain a highly-enriched population of viable B cells from splenocyte samples; using Akadeum’s Mouse B Cell Isolation Kit delivered an overall B cell purity of >92%, whereas they were only able to obtain 85% with a magnetic bead-based isolation kit. In addition to providing excellent purity, the gentleness of the microbubble isolation produced unblemished B cells that retained their antigen presenting potential.
Differentiation Potential Maintained Through Microbubble-Based Separation
A research group at the University of Pennsylvania used the Mouse B Cell Isolation Kit from Akadeum Life Sciences in their research into the mechanisms that underlie the differentiation of activated B cells into antibody-producing plasma cells. The sample preparation using Akadeum’s Mouse B Cell Isolation Kit delivered a highly-enriched sample with B cell purity of greater than 97%. Following plasma cell induction, upregulation of CD138 was confirmed in the enriched B cell population, validating post-enrichment function.
Akadeum’s Mouse B Cell Isolation Kit delivers a high-purity population of mouse B cells that are ready for downstream use. Use of these microbubbles to enrich mouse splenocyte samples for B cells prior to flow sorting can greatly reduce final sort times, helping to isolate more cells for better science. Akadeum’s microbubble workflow is exceptionally gentle on cells, helping to maintain the health and physiology of immune cells during enrichment. This even allows for the separation of fragile mouse naïve cells.
How it Works: First, Akadeum’s biotinylated antibody cocktail is added to the sample to label the contaminating cells. Next, the microbubbles are mixed into the sample where the seek out and bind to the unwanted cells. Once bound, they quickly and gently float the undesired cells to the surface. An optional centrifugation step pellets the B cells, and the microbubbles, microbubble-bound cells, and supernatant are easily aspirated away leaving the untouched and enriched B cells happy, healthy, and ready for downstream use.
Akadeum’s Mouse B Cell Isolation Kit delivers a highly enriched population of healthy, viable B cells using a protocol that is fast, easy, and gentle on cells–as does our Human B Cell Isolation Kit. Ideal for enrichment of delicate immune cells, Akadeum’s novel Buoyancy Activated Cell Separation (BACS™) occurs directly in the sample container without exposing cells to harsh chemicals or harmful shear forces.
The Akadeum microbubble enrichment protocol enables quick and easy sample preparation that takes about 30 minutes from start to finish without additional equipment or expensive consumables like magnets or columns. The end result is a high-purity population of mouse B cells that are ready for downstream use – in fact, using Akadeum’s Mouse B Cell kit prior to cell sorting can greatly reduce sort times, saving time and money while delivering a highly viable final population.
