Enhancing T Cell Isolation with Akadeum’s Alerion™ Microbubble Cell Separation System: A Case Study With Comprehensive Cell Solutions

Comprehensive Cell Solutions (CCS), a business unit of the New York Blood Center Enterprises, is a Contract Development and Manufacturing Organization. CCS recently evaluated the Alerion Microbubble Cell Separation System, the BioRise consumable, and the negative selection Human T Cell Leukopak Isolation Kit. CCS is dedicated to streamlining their manufacturing process and requires a platform that is flexible, efficient, and consistently delivers high-performance results to meet the demands of cell therapy manufacturing and development.

Kit Used

The Alerion™ Microbubble Cell Separation System was paired with Akadeum’s Human T Cell Leukopak Isolation Kit and BioRise consumable.

Experimental Design

To conduct this evaluation, CCS performed a split-run experiment with apheresis material from two healthy donors. Each leukopak was washed following CCS’s standard washing procedure and then split in half, approximately 7 billion cells in each half. Each half from both donors was then processed using the Alerion Microbubble Cell Separation System and the BioRise consumable in combination with Akadeum’s negative selection Human T Cell Leukopak Isolation Kit or a standard positive selection magnetic-based kit and corresponding instrument.

Discussion

The Alerion Microbubble Cell Separation System and the BioRise consumable, combined with Akadeum’s negative selection Human T Cell Leukopak Isolation Kit, provide unmatched results. 

  • Enhanced impurity depletion: Akadeum’s platform depletes B cells and monocytes, two cell types that can hinder T cell expansion and transduction, more effectively compared to the conventional magnetic platform.
  • Greater viability: The Alerion Microbubble Cell Separation System isolated samples maintained over 12% greater viability for each donor after cryopreservation than the conventional magnetic platform. When developing cell therapies, preserving cell health throughout processing is critical for efficacy. 

Conclusion

Akadeum’s Alerion™ Microbubble Cell Separation System, the BioRise™ consumable, and the negative selection Human T Cell Leukopak Isolation Kit provided CCS with enhanced results compared to the conventional magnetic method. The Alerion™ was able to process faster and provide more pure T cell populations while also maintaining cell health. Ultimately, producing a higher-quality intermediate, giving the potential for a better-quality product downstream.
