Enriching CD3+ T Cells, Allerion Case Study

A cell therapy company sought to improve its CD3+ T cell isolation process. To address the challenges it faces with its current method for recovery, purity, and processing time, it partnered with Akadeum Life Sciences to evaluate the Alerion™ Microbubble Cell Separation System in conjunction with the BioRise™ consumable and Human T Cell Leukopak Isolation Kit.

Objective

The primary objective was to evaluate the effectiveness of the Alerion™ Microbubble Cell Separation System for enriching CD3+ T cells from apheresis samples. The focus was on achieving highly pure and viable CD3+ T cells post-isolation.

Kit Used

The Alerion™ Microbubble Cell Separation System and BioRise™ consumable were paired with Akadeum’s Human T Cell Leukopak Isolation Kit.

Experimental Overview

The study isolated T cells from washed apheresis material using the Alerion™ Microbubble Cell Separation System. Key performance metrics evaluated included the input and output percentages of CD3+ T cells and post-enrichment viability.

  1. Sample preparation: Apheresis samples were obtained and processed to remove red blood cells and platelets, resulting in washed apheresis material.
  2. Isolation and cell counting: A sample was taken and read on a NucleoCounter® NC-200™ cell counter to determine cell counts. The rest of the washed apheresis material was processed using the Alerion™ Microbubble Cell Separation System. T cells were enriched via the BioRise™ consumable paired with the Akadeum Human T Cell Leukopak Isolation Kit. The resulting enriched cells were then counted and assessed for viability.

Key Findings

Enhanced Purity and Recovery for Better Cell Therapy Workflows

The Alerion™ Microbubble Cell Separation System, in combination with the Human  T  Cell Leukopak Isolation Kit and BioRise™ consumable, resulted in a 96.8% pure T cell population compared to the input material, which was at 34.6%.

This high output purity indicates that the Alerion™ Microbubble Cell Separation System effectively enriches CD3+ T cells for further therapeutic applications. Improved purity and recovery are critical for more efficient engineering of T cells, which in turn can enable better therapeutics.

High Viability, Better Cell Health

The viability of the CD3+ T cells post-enrichment was measured at 97.8%, demonstrating that the Alerion™ Microbubble Cell Separation System enriches the T cells without jeopardizing cell health. The efficiency and gentleness at which the Alerion™ system separates cells contribute to cell health. Healthier cells are better-engineered cells.
